Cell culture and reagents HIBEC lines and a human monocytic leukemia cell line (THP-1) were obtained from the Center of Hepato-Pancreato-Biliary Surgery in the First Affiliated Hospital of Sun Yat-sen University. Both HIBECs and THP-1 cells were cultured in RPMI-1640 medium (Gibco, Carlsbad, USA) supplemented with 10% fetal bovine serum (Gibco, USA) with 5% CO 2 at 37 °C. THP-1 cells were treated with 200 ng/mL of phorbol-12-myristate-13-acetate (PMA) (HY-18739; MedChemExpress, USA) for 48 h and then treated with 20 ng/mL of IL-4 (NBP2–35131; Novus, USA) to generate M2 polarization or treated with 100 ng/mL of lipopolysaccharide (HY-D1056; MedChemExpress) to generate M1 polarization. Recombinant Maltose Binding Protein (MBP) and MBP- Cs GRN were obtained from our previous study [14]. The Cs GRN fragment was amplified using forward primers 5′-ATA AGG ATC CCG GAG CAC AGGTGTAG-3′ (EcoRI) and reverse primers 5′-CGC GGA TCC TGT AAA TATA ACC AGA

Further data on Paxlovid's therapeutic potential in Covid-19 is likely to be released in the coming years. Credit: creativeneko via Shutterstock. At the 33rd European Congress of Clinical Microbiology and Infectious Diseases (ECCMID) 2023 conference in Copenhagen, Denmark, a year-long review of infectious diseases discussed some of the most important findings of 2022, relating to viral, bacterial, and fungal infections. More than two years after the start of the pandemic, Covid-19 remained a key area of research interest throughout 2022. A retrospective study provided the first real-world data on the use of antiviral therapy Paxlovid (N-Rtv) to treat Covid-19 and its impact on hospitalisations among adult patients between March and August 2022. Decreases in hospitalisati

Dog owner warns of rare fungal infection found in pets - WWNY

Image
OSHKOSH, Wis. (WBAY/Gray News) – A woman in Wisconsin says her two dogs have been battling a blastomycosis infection for over a year and wants to share her story so other dog owners know what signs to look for. Blastomycosis is an uncommon but potentially serious fungal infection that primarily affects the lungs. Lisa Kavanaugh said her dogs, Zena and Conan, have both been fighting the infection since December 2021. She started noticing symptoms in Zena first. She was lethargic, didn't want to eat and had swollen lymph nodes, as well as other symptoms that resembled pneumonia. "Symptoms are usually a draining track like a wound that won't heal. They can present with it in their bone, show signs of limping. Coughing is the most common because they inhale in their nose, so lung symptoms, it would look like pneumonia," veterinary Dr. Jamie Zarda explained. It just so happens Kavanaugh lost a dog in the 90s to blastomycosis, so she asked her vet to test Zena. She was posi

Covid Long Haulers Show Us How to Transform Healthcare - Psychology Today

Image
On a cold night in February 2021, I lay in the dark in my parents' basement, worried that illness might be my permanent future. It was months after Covid struck me and stayed with me. The disease felt like a phantom demon was clawing out my nervous system. Each night my 69-year-old mother gave me a massage. It felt like she was squeezing energy back into my dying muscles. Just over a year earlier, I had written the story with the highest reader engagement of any story CNN published in 2019. And it came during a big year for the biggest online news outlet in the world. Buoyed by a string of chart-topping stories, I had plunged directly into covering the pandemic. It felt like the most exhilarating time in history to be a science writer. Then, nine months in, the virus got to me. I still wasn't better months after I had tested positive. Long Covid is often discussed as fatigue and brain fog. However, it's better described as a total deadness of

Strep throat is back; antibiotic suppliers are working to meet demand - The Washington Post

Even as the winter's respiratory virus season fades, strep throat infections remain high, and in some cases pediatric formulations of some antibiotics are in short supply. A more severe form of strep can infect the bloodstream and has also increased in children and adults, according to the Centers for Disease Control and Prevention. We spoke to infectious-disease experts to answer common questions about strep, its symptoms and the antibiotic shortages affecting some parts of the country. What to know Adblock test (Why?)

A Parasite Is Killing Sea Otters. Is Cat Poop to Blame? - WIRED

Image
This story originally appeared in The Guardian and is part of the Climate Desk collaboration. Scientist Melissa Miller was seeing something in California sea otters that she had not seen before: an unusually severe form of toxoplasmosis, which officials have confirmed has killed at least four of the animals. "We wanted to get the word out. We're seeing something we haven't seen before, we want people to know about it and we want people working on marine mammals to be aware of these weird findings," said Miller, a wildlife veterinarian specialist with the California Department of Fish and Wildlife (DFW). "Take extra precautions." In March, a study from the DFW and the University of California, Davis, revealed that a rare strain of the parasite, never before reported in aquatic animals, was tied to the deaths of four sea otters. The strain, first seen in Canadian mountain lions in 1995, had not been previously detected on the California coast. "This w

An Infection Preventionist's Role in Dentistry: What Changes Need ... - Infection Control Today

Image
How is infection prevention different in dental offices from other health care facilities? Experts explain. Infection prevention (IP) doesn't stop at the hospital door. One place where IP may be overlooked is in the dentist's office. Infection Control Today ( ICT ) spoke to Amanda Hill BSDH, RDH, a registered dental hygienist and industry consultant, and Michelle Strange, MSDH, RDH, CDIPC, owner of Level Up Infection Prevention (www.levelupip.com) about why IP in dentistry is essential. ICT: Why is IP important in dentistry? Michelle Strange, MSDH, RDH, CDIPC : We all learn a little bit about IP and think we know it all. The industry is self-regulated, which is a bit dangerous, and…we all get very comfortable in our procedures and behaviors. We need to remember…how critical patient safety is and that we are doing these minisurgical procedures regularly. We are managing diseases in people's mouths, periodontitis, wound management, [and] surgical extractions, and placing imp

Bird shots: Is vaccinating poultry the best defense against a deadly ... - Science

Lakeside, California— Hilliker's Ranch Fresh Eggs in this San Diego suburb has 30,000 chickens in three "cage-free," open-air barns, where birds crowd the floor like rush-hour riders on a big city subway. "A cage-free aviary is a very interesting science experiment," says Frank Hilliker, who runs the farm his grandfather started in 1942. He worries mightily about infections spreading through the massed birds. On his iPhone, he pulls up a list of the vaccines his chickens get: against Newcastle disease, infectious laryngotracheitis, coryza, colibacillosis, salmonella, infectious bronchitis, and fowlpox. There is one disease for which his chickens have not been vaccinated: a highly pathogenic avian influenza (HPAI) that's now racing around the world, killing 90% or more of the poultry it infects. Endemic in migratory birds, this HPAI emerged in U.S. poultry in February 2022, and to date has killed or required the culling of a record 58 million chickens, turk

Charlie Crist during his election night watch party at the Tampa Convention Center on November 8, 2022 in Tampa, Florida.
